New Process for Cell Detachment: Use of Heparin

Popis: The use of animal derived products like trypsine or serum presents a risk due to the potential transmission of adventitious agents. Their replacement by non animal derived or pharmaceutical grade substances is the best choice for avoiding this risk. Trypsin, a widely used substance for the detachment of anchorage dependent cells, can be replaced by heparin in most of the cases. At a concentration of 100IU/ml in the presence of EDTA at 0.02% in PBS. 75% (9/12) of all cell lines tested could be detached from polystyrene based growth supports (T-flasks, roller bottles, the Costar CellCube) without the need of optimizing the detachment conditions. These so detached cells could easily be subcultivated whereby a washing step for eliminating the detachment solution was preferable. The comparison of the growth rates of these subcultures revealed only insignificant differences for cells detached by trypsin/ EDTA, heparin/EDTA or EDTA alone (e.g. for BHK21 C13, recombinant CHO and CRIP M11 cells grown on polystyrene surfaces).
