Popis: | The strainStreptomyces aureofaciens Bg producing chlortetracycline, was grown in submerged cultures in flasks on a shaker, without maintaining aseptic conditions. If the medium was inoculated with a vegetative inoculum at least 24 hours old and containing at least 200 (μg./ml. of chlortetracycline, contamination of the cultures did not occur and the production of chlortetracycline was not reduced as compared with control fermentation carried out under the usual conditions of asepsis. Experimental contamination with bacteria, yeasts and natural material such as highly contaminated river water or soil did not cause any reduction in the production of chlortetracycline, with the exception of yeasts of the genusTorilopsis. Gram-negative rods resistant to chlortetracycline were isolated from cultures of the strainS. aureofaciens Bg contaminated with soil and with river water; these slightly stimulated production of the antibiotic if injected into the medium together with the production strain. Only yeasts of the genusTorulopsis, in massive experimental contamination, were capable of growth in a medium inoculated with the strainS. aureofaciens and reduced the production of chlortetracycline. These experiments demonstrate that strict observance of asepsis is unnecessary in the fermentation production of chlortetracycline. |
