Pseudomonas mangiferae sp. nov., isolated from bark of mango tree in Thailand

Popis: An aerobic, Gram-stain-negative, rod-shaped bacterium, designated DMKU BBB3-04T, was isolated from bark of mango tree . Colonies were circular, convex with entire margins when grown on nutrient agar medium for 2 days. The bacterium was motile by means of lophotrichous flagella and produced black-brown pigment. The strain grew at 12–40 °C (optimum, 28–30 °C) and at pH 6.0–8.0 (pH 6.5). Growth was observed in the presence of up to 5 % (w/v) NaCl. Strain DMKU BBB3-04T showed the highest 16S rRNA gene sequence similarity to Pseudomonas furukawaii KF707T (97.1 %), Pseudomonas indica NBRC 103045T (97.0 %), Pseudomonas tarimensis MA-69T (96.9 %) and Pseudomonas azotifigens 6H33bT (96.8 %), suggesting that strain DMKU BBB3-04T should be classified within the genus Pseudomonas . Analysis of strain DMKU BBB3-04T was also performed using three housekeeping genes (gyrB,rpoB and rpoD) and further confirmed the phylogenetic assignment of the strain. The major fatty acids found in strain DMKU BBB3-04T were summed feature 8 (C18 : 1 ω7c/C18 : 1 ω6c), C16 : 0 and summed feature 3 (C16 : 1 ω7c/C16 : 1 ω6c) (35, 22 and 19 %, respectively). The major polar lipids consisted of diphosphatidylglycerol, phosphatidylethanolamine and phosphatidylglycerol. The respiratory quinone was Q-9. The DNA G+C content of the strain DMKU BBB3-04T was 67.6 mol%. On the basis of phenotypic characteristics, chemotaxonomic data and phylogenetic analysis, strain DMKU BBB3-04T is considered to represent a novel species in the genus Pseudomonas . We propose the name Pseudomonas mangiferae sp. nov. for this novel species. The type strain of the novel species is DMKU BBB3-04T (=TBRC 7080T=JCM 32061T).
