Popis: With burgeoning research in biomass to fuel industry, the world has seen limited success in implementing second generation, as well as advanced biofuels. The key here is to release fermentable sugars from the lignocellulosic biomass in a cost-effective manner. This requires identifying and benchmarking biomass degrading enzymes namely cellulases from different commercial/engineered/secretome preparations and designing potent cocktail. However, one of the hurdles with commercial and/or secretome preparations is that the majority of them have undefined and unknown compositions and hence it becomes difficult to benchmark such preparations. Likewise, the knowledge pertaining to the benchmarking and characterization of the cellulase system is limited to individual application and needs to be revisited for their holistic understanding. In this chapter, we have attempted to streamline the application based important enzyme characteristics such as biochemical analysis, structural features and associated mechanistic behavior. This chapter also highlights, use of existing enzyme assays and new-generation analytical equipment to characterize cellulases for its application in biomass conversion.
