The stimulation of respiration by uncoupling agents, called respiratory control by analogy with the stimulation of respiration by ADP plus phosphate, has been very useful in studies of the mechanism of energy coupling in oxidative phosphorylation. LOOMIS and LIPMANN (1948) originally reported that the rate of respiration of isolated mitochondria is increased by addition of 2,4-dinitrophenol. This led to the concept that such “uncoupling” agents break down a high-energy intermediate and thus relieve the back pressure of the coupling reactions on the oxidation reactions of the respiratory chain (SLATER, 1953). LEE and ERNSTER (1966) extended these observations by demonstrating that sonically fragmented vesicles of mitochondria, submitochondrial particles, also showed a stimulation of respiration by uncoupling agents, and that the controlled respiration of such preparation corresponded to the ability to couple respiration to various energy-linked functions. |
