Popis: | Male rats (Sprague-Dawley) were sacrificed when 12 weeks of age. Aliquots (0.4 gm) of a testicular mince from these animals were incubated and irradiated simultaneously for 15 minutes at 37.5°C in 10 ml of phosphate buffer, pH 7.4, containing 2.275 mμmoles of $\text{pregnenolone-}7\alpha \text{-}{\rm H}^{3}$ and $\text{progesterone-}4\text{-}{\rm C}^{14}$ . One group of samples was exposed to 858 R of X-rays, while a second group was exposed to 4050 R during the 15-minute incubation period. Sham-irradiated control samples were irradiated and incubated with boiled testicular mince to ascertain if irradiation per se would decompose the radioactive steroid substrates. Irradiation of testicular mince with 858 R resulted in an increased androgen production. This increased androgen production was accompanied by a decreased 17α-hydroxylation and a decreased synthesis of compounds with a polarity greater than testosterone. At the higher dose, androgen synthesis and 17α-hyd... |
