Bioinformatics Approaches for Analysing the Spatial Structure and B-cell Epitope of Eg14-3-3 Protein

Popis: | Echinococcus granulosus (E. granulosus) is a small parasitic flat worm that would cause a zoonosis named Cystic echinococcosis (CE), which leads to a serious healthy and economic damage all over the world. To prevent the spread of the disease, the vaccine against E. granulosus has become one of the most effective strategies nowadays. According to the previous research, Eg14-3-3 protein is an important vaccine candidate antigen against E. granulosus. Therefore, serval bioinformatics approaches were used to analyze the spatial structure and linear B-cell epitopes of Eg14-3-3 protein. The composition of secondary structural is 68.42% alpha helix, 7.69% extended strand, 0.81% beta turn and 23.08% random coil. The Phyre2 server was selected to construct the tertiary structure of Eg14-3-3. The BCPREDS Server 1.0 and ABCpred Prediction Server were chosen to predict the linear B-cell epitope. Comparing the result of two software, there are 5 dominate epitopes occurred: 66-71, 76-81, 121-127, 131-144, 227-232. Through the spatial structure and B-cell epitopes prediction, this study provides theoretical basis for the future vaccine against E. granulosus. |
