FL 1039: A new β-lactam derivative for the treatment of infections with gram-negative bacteria

Popis: | The efficacy of a new β-lactam antibiotic FL 1060, 6β-[(hexahydro-1-H-azepin-1-yl)-methyleneamino]-penicillanic acid, given orally as its pivaloyl-ester FL 1039, in eradicating Salmonella from chronic carriers and acutely ill patients was studied. Changes in the normal intestinal flora were also investigated. Out of 12 carriers eight were cured by FL 1039, in three cases in combination with cholecystectomy. In the four carriers not cured, resistant Salmonellae emerged (in three instances carrying R factor) but they became sensitive again within one month. In six acutely ill patients the period of illness was not shortened; on the other hand the excretion period of Salmonella was prolonged in one case only in comparison with control patients not receiving antibiotic therapy. In this case the Salmonella became resistant and R factor-carrying. — The influence of FL 1039 on the normal intestinal flora was moderate. No overgrowth of fungi, staphylococci or Pseudomonas was noticed. The emergence of resistant and R factor-carrying enteric bacteria was comparable to that following ampicillin treatment. |
