It has been suggested that endogenous adenosine protects neurones from the damage induced by such pathological conditions as ischemia, hypoxia and epilepsy (1). Adenosine has many effects on the central nervous system, several of which could be involved in neuroprotection. Adenosine can improve the cerebral blood supply (2), limit the energy needs of cerebral tissue by preserving ionic homeostasis and hyperpolarizing neurons (3), or block specific events such as release of excitatory amino acids (4) or free radical production (5) that trigger neuronal death. We have investigated the possible roles of adenosine in two experimental pathological conditions that elicit localized neuronal damage in the rat: long lasting seizures induced by kainic acid (KA) and transient forebrain ischemia. Two strategies were used: blocking adenosine receptors with theophylline in KA-induced seizures, and enhancing the effects of adenosine with R-phenyl-isopropyl adenosine (R-PIA) in forebrain ischemia. |
