Recombinant soluble human α1,3/4FucosyltransferaseIII was produced with two Sf9 insect cell lines adapted to grow either in serum-containing (SCM) or in serum-free medium (SFM). The optimal time of infection to obtain the highest Fuc-TIII activity in the supernatant was mid-exponential growth phase in SCM, whereas in the SFM better results were obtained by infecting cells at the end of the exponential growth phase. The enzyme produced in the absence of serum was more stable when stored at 27°C and 4°C. Western-blot analysis of the two proteins revealed a higher heterogeneity and a slightly inferior molecular weight for Fuc-TIII produced in SFM. |
