Popis: | Salt content of basal modified cellobiose-polymyxin B-colistin (mCPC) medium was varied to assess impedance-based detection of Vibrio cholerae and Vibrio vulnificus. Several sodium salts were tested as substitutes for NaCl. Bactometer detection times (h) were compared to find the optimal salt and its concentration. Results using tryptic soy broth indicated 2% Na2HPO4 was the best NaCl replacement. Use of 2% Na2HPO4 with 0.5% NaCl in mCPC medium allowed rapid detection of both vibrios using a capacitance test code. Correlation of capacitance detection times with MPN/ml gave r = −0.74 and r2 = 0.54 for V. cholerae and r = −0.68 and r2 = 0.46 for V. vulnificus. With starting counts above 104 MPN/ml, both pathogens could be enumerated in less than 20 h with this method. |
