Amplification of extracted nucleic acids is often necessary to facilitate analysis of their nucleotide sequences or expression patterns. In this section, we review the most commonly used amplification method in molecular laboratories worldwide, the polymerase chain reaction (PCR). Additionally, PCR primer design, optimization techniques, and contamination prevention are discussed. Finally, many of the most commonly used PCR variations (e.g., multiplex PCR, reverse transcription-PCR, and quantitative PCR) are also described. |
