Crystallization and Diffraction Studies of Thermus Flavus 5S rRNA and Synthetic Fragments of the 5S rRNA

Popis: Although it has been established that the ribosomal 5S RNA is essential for the biological function of the ribosomes (Nomura and Erdmann, 1970; Erdmann et al., 1971 a, b; Nierhaus and Dohme, 1974; Dohme and Nierhaus, 1976; Hartmann et al., 1988), its precise role in protein synthesis remains to be elucidated. This dilemma is partially due to the fact that its structure in the ribosome and how it interacts with the ribosomal proteins is still unknown. Since the numerous chemical, biochemical and computer aided studies were insufficient in describing the three-dimensional structure of this molecule (Wolters and Erdmann, 1988; Lorenz et al., 1989; Specht et al., 1991; Bald et al., 1992), we are attempting its crystallization. In this communication we will summarize our efforts to crystallize the ribosomal 5S RNAs, their protein complexes and chemically synthesized domains of Thermus flavus 5S rRNA.
