The allograft response represents an exceedingly complex phenomenon that involves multiple cellular and soluble mediators. Of central importance to the initiation as well as perpetuation of this response is the recognition by recipient T cells of donor-derived major histocompatibility complex (MHC) antigens expressed on the transplanted tissue. The recognition of allogeneic MHC antigens by recipient T cells is mediated by clonally restricted T-cell receptors (TcR) in conjunction with nonpolymorphic accessory molecules that regulate adhesion, signal transduction, and activation of the responding celU The TcR provides specificity to the response by interacting with the polymorphic regions of the MHC and, along with the associated CD3 complex, mediates cellular activation. |
