Popis: Stable expression of human Insulin-like growth factor binding protein-1 (IGFBP-1), as determined by a biosensor, the BIACore, at high levels has been achieved in Chinese Hamster Ovary (CHO) cells by cotransfection and subsequent coamplification of expression vectors containing the IGFBP-1 cDNA and a dihydrofolate reductase (DHFR) cDNA gene into CHO DHFR-deficient cells. Stepwise selection of the DHFR+ transformants in increasing concentrations of methotrexate generated cells with a high copy number of IGFBP-1 genes. IGFBP-1 expression was found to increase withincreasing copy number. These cells werestably amplified and expressed high levels ofhuman IGFBP-1 in the absence of MTX. Aflow cytometric method was used to measure the intracellular level of IGFBP-1.
