Quantification of the hypothalamic proline rich polypeptide-1 in rat blood serum

Popis: Proline rich polypeptide-1 (PRP-1) consisting of 15 amino acid residues (AGAPEPAEPAQPGVY) is a regulatory hypothalamic peptide possessing neuroprotective, immunoregulatory, hematopoietic, and antimicrobial properties. In order to quantify PRP-1 in rat blood, an enzyme linked immunosorbent assay (ELISA) has been applied using polyclonal antibody raised against the synthetic PRP-1 named “Galarmin”. In the present study, two post-injection periods were tested (5 hours and 2 days) after intraperitoneal (i/p) administration of PRP-1. The minimum detectable concentration of PRP-1 in intact rat blood serum has been shown to be approximately 1.78 ng/mL. A significant increase of PRP-1 concentration in the blood samples has been observed in 5 hours after the PRP-1 i/p injection; however the peptide amount in the blood has been decreasing in 2 day post-injection period, approximating to the control level, which could be caused by the peptide proteolitic breakdown.
