In order to quantitatively analyze γ-glutamyl-e-lysine, [G-3H]dansyl chloride was used to label [U-14C]leucine and the isopeptide. After thin-layer chromatography of the products, the dansylated amino acid and isopeptide were acid hydrolyzed for up to 18 hr. The tritium was found to be labile under these conditions, with a half-life of exchange of about 5 hr, and the system behaved as if two labile tritium atoms were exchanging with pseude-first-order rate constants of 0.5 and 0.05 hr−1. The errors introduced by acid hydrolysis render this reagent unsuitable for quantitative analysis. |
