Popis: SummaryThe commercial propagation of date palm is through offshoots only, although these species can be multiplied by seeds also. Date palm being dioecious tree, the seed progeny, however, would have large number of male plants and the female plants too may be genetically heterogenous. This necessitates the propagation of date palm through offshoots. Unfortunately, number of offshooys in mother palm trees is also limited. Use of plant tissue culture techniques for fast multiplication of date palm, therefore is being considered by several tissue culture laboratories.A method has been developed to regenerate large number of plants from callus established from axillary buds and shoot tips of 3–4 year old offshoots in two varieties of date palm—Medjool and Khadravi. Best results were achieved when these ex-plants were cultured on a modified MS medium containing activated charcoal (0.3 %), Na H2PO4.2H2O (170 mg/l), KH2PO4.2H2O (200 mg/l), 2,4-D (100 mg/l), BAP (5 mg/l) and thiamine Hcl (1 mg/l).
