Bis-ANS is a marker of hydrophobic sites. Its interaction with bovine α-lactalbumin (BLA) is studied by fluorescence titrations of 1 μM bis-ANS with BLA as well as by titrations of 1 μM BLA with bis-ANS. To reduce inner filter effects at high probe concentrations, an excitation wavelength of 455 nm, on the edge of the bis-ANS absorption peak, has been chosen. Even at 600 μM bis-ANS, the absorbance does not exceed 0.3. The fluorescence emission between 515 and 550 nm has been integrated, corrected for inner filter effects and the background fluorescence has been substracted. |
