Identification and mutagenicity of dinitrotoluene metabolites produced by strain Pseudomonas putida OU83

Popis: | Pseudomonas putida OU83 was isolated from soil contaminated with substituted aromatics. Resting cells of this bacterium incubated with dinitrotoluenes (DNTs) produced potentially biohazardous biotransformation products. The GC–MS analysis of the metabolites revealed fragmentation pattern of molecular ions at 152, 135, 107, and 77 amu, suggesting formation of methylnitroanilines. The identification of the metabolites was confirmed by the NMR spectral analysis. These results showed that 2,4-DNT, 3,4-DNT, and 2,6-DNT were transformed to 4-methyl-3-nitroaniline, 4-methyl-2-nitroaniline, and 2-methyl-3-nitroaniline, respectively. Relative biotransformations of 2,4-DNT, 3,4-DNT, and 2,6-DNT were to be 98%, 75%, and 40%, respectively in 48 h. These results indicate that strain OU83 enzymes catalyze nitro group of the DNT to an amino group and can be used for commercial production of methylnitroanilines, an intermediate for dye manufacturing. The mutagenicity of the metabolites of DNTs was increased, as tested by using Salmonella typhimurium strains TA98 in presence but not in the absence of arochlor 1254-induced (S9) rat liver mix. These findings suggest that the metabolites of DNTs produced by P. putida OU83 are promutagens. |
