Regulation of miR101/miR‐199a‐3p targeting cyclooxygenase‐2 by the epithelial sodium channel during embryo implantation (911.5)

Popis: Decidualization is a maternal prerequisite for embryo implantation. However, it remains unclear how signals from the embryo at the endometrial surface are converted into chemical signals, leading to decidualization in the stromal cells. Our previous study has demonstrated that the epithelial sodium channel (ENaC) plays a critical role in embryo implantation by regulating the decidulization-inducing molecules cyclooxygenase-2 (COX-2)/PGE2. Recently, we also found that ENaC activation in human endometrial cells could triggers Ca2+ release, phosphorylation of cAMP response element-binding protein (CREB), down-regulation of mir101/mir199a-3p with up-regulation of COX-2. In line with in vitro data, knocking down CREB in mice uterine resulted in mir101 and mir199a-3p up-regulation, COX-2 down-regulation and implantation failure. Taken together, these results indicate that ENaC-dependent activation of CREB may suppress mir101/mir199a-3p, resulting in up-regulation of COX-2 necessary for embryo implantation. The ...
