Popis: | A colorimetric method is described for the estimation of the amount of protein in the growth hormone and prolactin bands of disc electrophoretic patterns. After separation by electrophoresis in an alkali-soluble polyacrylamide gel, a band of stained hormone is cut from the gel, dissolved, and the optical density of the solution measured. The optical density value is converted to micrograms of protein by reference to a standard curve made with a highly purified preparation of the hormone. There was excellent correlation between biological potency and the optical density value obtained with a given weight of a purified hormone. The concentrations of growth hormone and prolactin in the pituitary gland of the rat and mouse as determined by the electrophoretic method are reported, and, although bioassays on the glands were not carried out in this study, the values agreed quite well with those that have been reported by a number of investigators.
