Omeprazole, an Inhibitor of H+-K+-ATPase, Markedly Reduces CSF Formation in the Rabbit

Popis: CSF formation can be inhibited by ouabain, suggesting an involvement of transport systems dependent on sodium-potassium activated adenosine triphosphatase (Na+-K+-ATPase) (Cserr 1975). Recently, it has been found that the gastric mucosa of pig, dog and guinea-pig contains another specific ATPase, which is H+-K+ dependent (Saccomani et al. 1979). Inhibition of this enzyme with omeprazole, a substituted benzimidazole, markedly and selectively reduces histamineand db-cAMP-stimulated gastric acid secretion (Wallmark et al. 1983). In view of similarities between the secretory responses in the gastric mucosa and choroid plexus epithelia to drugs affecting the autonomic nervous sytem, and since the choroid plexus seems to playa role in the regulation of the pH in the CSF, it was considered of interest to determine whether H+ -K+-ATPase-like activity is present in the choroid plexus by measuring the effect of the selective inhibitor, omeprazole, on choroid plexus ATPase activity, transport function, and rate of CSF formation.
