Application of a flow system with laser polarimetric automatic detection for molecular exclusion chromatography of carbohydrates

Popis: The combination of molecular exclusion liquid chromatography and laser polarimetric detection has turned into a carbohydrate separation and quantification system for plants fluids of industrial value, opening the possibility to evaluate, among others, the quality of sugarcane juices and agave extracts. Some previous papers described a system where liquid chromatographic carbohydrate separation and polarimetric detection by means of a Laserpol 101-M was used for discrete fractions. In this paper, a new improved system has been discussed which accomplishes polarimetric measurements of eluates in a continuous flux directly from a liquid chromatographic system, giving also the possibility to isolate fractions of interest. Chromatograms of sucrose, fructose, glucose and their mixtures with dextran obtained under both regimes of work are shown to compare the accuracy of continuous flow system. Outstanding results were obtained when variations in carbohydrate composition occurring in sugarcane plants affected by leaf scald were submitted to study at ages when visual symptoms are not apparent. A computer program allowing the output of chromatograms to a display on line and the capability of digital data storing, maximum detection, zone integration, and some other possibilities make this new system very competitive and self-convincing.
