An original method is presented, allowing the estimation in urinary extracts of 5 to 100 μg of pregnanediol, in 4 h and with a high specificity. It involves rapid hydrolysis of Helix pomatia with β-glucuronidase and sulfatase and a single extraction with a non-toxic, non-emulsive, little inflammable solvent mixture, yielding very few pigments in the extracts. The final assay is performed by gas phase chromatography on trimethylsilyl derivatives. Sensitivity, specificity, accuracy and reliability of the method are studied. |
