RNA polymerase-DNA complexes resistant to deoxyribonuclease digestion (“kegs”) and purified keg DNA were isolated, and their properties with respect to the binding of actinomycin D were studied by spectrophotometric methods. Actinomycin D binds to the kegs but does not cause dissociation of the RNA polymerase-DNA complex. The apparent number of binding sites in the kegs is approximately equal to the value obtained for the binding of actinomycin D to free keg DNA. The apparent association constant calculated from the binding of actinomycin D to the kegs differs markedly from that observed for the binding to free keg DNA (or total calf thymus DNA). This may reflect important structural differences between free DNA and DNA complexed with RNA polymerase. |
