The interaction of a nonionic detergent with protein

Popis: The titration curve of bovine plasma albumin showed a slight shift in the acidity constants of the carboxyl groups to higher values in the presence of the polyxyethylene nonionic detergent, Triton X-100. The p K a of the dimethylamino group of an azomercurial linked to protein is lowered by addition of Triton X-100 to the solvent. This change is accompanied by a blue shift in the absorption peak of the dye, a shift similar to that observed when such dyes are transferred from an aqueous to a non-aqueous solvent. The changes in p K a and absorption spectrum of the albumin-dye complex are reversed, in part, by the further addition of 8 M urea. These data are consistent with the view that the detergent interacts with the protein reinforcing and enlarging the envelope of structured water around the protein.
