MicroRNAs (miRNAs) have been shown to control virtually all physio-pathological aspects of mammalian brain. Uncovering dynamics of miRNA expression and functions in brain health and disease are amongst the main challenges in modern neuroscience and purpose of pharmacological and technological R&D. This chapter lays out the different methodological approaches that allow the identification of novel miRNAs, expression analysis and the functional validation of miRNA–mRNA target cross-regulations in brain. The approaches described in this chapter are not self-standing alternatives to investigate miRNAs, but they are concurrent steps that converge to the investigation purpose. On the other hand, technological developments have been propellers of novel break-through discoveries in the neuroscience field, such as the identification of the extreme heterogeneity of brain cells and novel therapeutic applications, such as the use of miRNAs as next-generation of diagnostics and therapeutics. |
