The principle of PACIA (particle counting immunoassay) is based on the agglutination of polystyrene particles 0.8 µm diameter, usually called ‘latex’. The novelty of the method consists in the measurement of agglutination by determination of the number of residual non-agglutinated particles by means of a Technicon AutoCounter, an instrument designed for the counting of blood cells1. The advantages of the method are: (1) radioisotopes are not used; (2) reagent stability; (3) analysis rate of 60/h; (4) no separation step is required — that is, the assay is completely homogeneous; (5) all assays are completed within 30 min; (6) the sensitivity of the technique equals or exceeds that of RIA; (7) probably all determinations possible with RIA are equally possible with PACIA. |
