Popis: In cases of massive soft tissue damage, treatments for long-term repair and especially volume reconstitution are sought after. In such cases, tissue engineering could provide a valuable option for treatment. Since the degradation of many materials is still a constraint for the use of engineered constructs, the aim of this study was to develop a carrier which allows a more predictable and stable outcome. To this aim a safe biological carrier composed of a human allogeneic acellular dermis sterilised with a validated virus-inactivation method was produced and its suitability was assessed in terms of growth promoting effects and stability. Growth kinectics using human dermal fibroblasts cultivated on the material demonstrated a good cell proliferation. The high resistance to contraction by fibroblasts and collagenase digestion confirmed a high stability of the acellular dermis. Their good properties are mainly due to their biological origin containing highly crosslinked collagen in a natural supra-organisation. Populated collagen gels, composed of non-crosslinked collagen, were assessed in parallel and exhibited both lower proliferation and stability. Since human allogeneic virus-inactivated acellular dermis is very stable and supports cell proliferation, it could be a valuable carrier for tissue engineering offering the option of long-term tissue repair.
