Regulation of Thymic Lymphopoiesis in Vitro

Popis: The fundamental mechanisms involved in the regulation of thymic lymphopoiesis are largely unknown, including the properties of the lymphoid precursor cells and the factors that determine lymphoid cell proliferation and differentiation. We have therefore attempted to analyze possible regulatory mechanisms acting in thymic lymphopoiesis. Our experimental model has been the embryonic thymus in organ culture. In this system lymphoid cell development may be influenced both by the thymic microenvironment, including locally produced thymic hormone(s), and by factors present in the serum supplement of the organ culture medium. We have thus demonstrated and characterized a factor in chicken (1,2) and mammalian sera (3) essential for chicken respectively mouse thymic lymphopoiesis in vitro. Other work suggested an internal control of lymphoid cell proliferation and development in the thymus (1,4).
