Talampicillin (phthalidyl D-α-aminobenzylpenicillinate) is hydrolysed in the body to ampicillin. Although ampicillin is well absorbed, Talampicillin is even better absorbed, as shown by Human volunteer studies. In cross-over studies using a capsule formulation, Talampicillin gave rise to higher serum concentrations of ampicillin than an equivalent dose of ampicillin itself. In 10 volunteers dosed with 250mg of ampicillin, the mean peak serum concentration was 2.4µg/ml and occurred at 2 hours after dosing. When a dose of Talampicillin, equivalent to 250mg of ampicillin was given, the mean peak serum concentration was 5µg/ml at 40 minutes after dosing. In later studies with a tablet formulation of Talampicillin equivalent to 169mg of ampicillin, essentially similar peak concentrations of ampicillin were obtained. Thus, in man, Talampicillin is rapidly absorbed and rapidly hydrolysed. |
