Ð Ñакже оÑенено влиÑние пÑоизводнÑÑ Ð¼Ð¾ÑÑолина на моделÑнÑе ионнÑе каналÑ, обÑазованнÑе гÑаÂмиÑидином Ð (ÐÑÐ). УÑÑановлено, в завиÑимоÑÑи Ð¾Ñ Ð¸Ð·Ð¼ÐµÐ½ÐµÐ½Ð¸Ñ Ð³ÑаниÑного поÑенÑиала палÑмиÑоилÑоÑÑаÑÐ¸Ð´Ð¸Ð»Ñ Ð¾Ð»Ð¸Ð½Ð¾Ð²ÑÑ Ð¼ÐµÐ¼Ð±Ñан, моÑÑолин-ÑодеÑжаÑие веÑеÂÑÑва можно ÑазделиÑÑ Ð½Ð° ÑÑи гÑÑппÑ: имеÑÑие ÑилÑное, ÑÑеднее и Ñлабое дейÑÑвие. ÐÑновной вклад в изменение гÑаниÑного поÑенÑиала мембÑÐ°Ð½Ñ Ð²Ð½Ð¾ÑÐ¸Ñ Ð´Ð¸Ð¿Ð¾Ð»ÑÐ½Ð°Ñ ÑоÑÑавлÑÑÑаÑ. ÐдиноÑнÑе каналÑ, обÑазованнÑе гÑамиÑидином Ð, иÑполÑзовали в каÑеÑÑве обÑекÑа, ÑÑвÑÑвиÑелÑного к Ð¸Ð·Ð¼ÐµÐ½ÐµÐ½Ð¸Ñ Ð´Ð¸Ð¿Ð¾Ð»Ñного поÑенÑиала мембÑанÑ. ÐолÑÑеннÑе ÑезÑлÑÑаÑÑ Ð¿Ð¾ÐºÐ°Ð·ÑваÑÑ, ÑÑо повÑÑение пÑоводимоÂÑÑи ÐÑÐ-каналов в пÑиÑÑÑÑÑвии CNV-3 ÑоÑÑавлÑÐµÑ 15%, в Ñо вÑемÑ, как CNV-58 не влиÑÐµÑ Ð½Ð° даннÑй паÑамеÑÑ. СÑÐ°Ð²Ð½Ð¸Ð²Ð°Ñ Ð¸Ð·Ð¼ÐµÐ½ÐµÐ½Ð¸Ñ Ð´Ð¸Ð¿Ð¾Ð»Ñного поÑенÂÑиала мембÑÐ°Ð½Ñ Ð¸ Ð¸Ð·Ð¼ÐµÐ½ÐµÐ½Ð¸Ñ Ð¿ÑоводимоÑÑи ÐÑÐ-каналов, можно заклÑÑиÑÑ, ÑÑо Ð¼ÐµÐ¶Ð´Ñ Ð½Ð¸Ð¼Ð¸ наблÑдаеÑÑÑ Ð¿ÑÑÐ¼Ð°Ñ ÐºÐ¾ÑÑелÑÑиÑ. Morpholine derivatives are widely used in manufacturing and agriculture, as well as in pharmacology. Multiple studies show the possibility of morpholine deÂrivatives to influence the functioning of enzymes and receptors, mainly associated with lipid membranes of cells of the central nervous system (CNS), which makes them promising in the treatment of neuropsychiatric diseases associated with imÂpaired enzymatic and receptor processes.  The aim of this work is to study the effect of morpholine derivatives on the physicochemical properties of lipid membranes. Using electrophysiological and fluorimetric measurements changes in the boundary and dipole potential of the membranes were performed. The effect of morpholine derivatives on model ion channels formed by gramicidin A (GrA) was also evaluated. It was found that, depending on the change in the boundary potential of palÂmitoyl phosphatidylcholine membranes, morpholine-containing substances can be divided into three groups: with strong, medium and weak effect. It was shown that the change in the boundary potential is due to modulation the dipole component.Single channels formed by GrA were used as a potential-sensitive probe. The results obtained show that the increase in the conductance of GrA channels in the presence of CNV-3 is about 15%, while CNV-58 does not affect this parameÂter. One can assume that change in the GrAâconductance correlates with the change in the dipole potential of membranes. |
