The cytotoxic potential of 28 molecules was determined, by a cell viability test, on a rat hepatocyte monolayer culture, on a porcine Leydig cell culture and on an epithelial cell line of CH0-K1 cells. In 65% of the cases, the results were identical in the three models. In spite of better sensitivity, the primary culture models did not show a better ability to detect specific toxics than the cell line. This study demonstrates that the use of organ-specific primary cell cultures does not appear justified if the toxicity is evaluated by an aspecific cytotoxicity test. It seems necessary to evaluate more specific cell function parameters to assess, in vitro, the toxicity of molecules. KEYWORDS: rat hepatocytes, pig Leydig cells, CH0-K1 cells, in vitro cytotoxicity |
