After developing an in vitro assay system, we found that fish stomach and intestinal homogenates as well as purified fish pepsin and trypsin effect significant degradation of human epidermal keratin. The activity of trypsin and pepsin of five different fish species was studied. All the fish species had pepsin pH optimum around pH 3.3–3.4 and trypsin pH optimum between pH 8.5 and 9.5. Boiled homogenates and boiled enzymes caused no degradation. Specific inhibitors of pepsin and trypsin significantly inhibited the keratin-degradation process. Intracutaneous injection of trypsin and pepsin induced inflammation. Repeated percutaneous application of stomach and intestinal homogenates and purified trypsin and pepsin caused a slight degradation of the keratin layer and a slight inflammation was also observed. These results together may explain irritative contact hand eczema following continuous handling of stomach and intestinal contents of fish. |
