This review focuses on how follicular dendritic cells (FDC) and associated antigens are involved in germinal center reactions and how changes in FDC function alter humoral immune responses. In normal animals, FDC trap antigen in the form of immune complexes and these antigens are presented to B cells, which then process and present the antigen obtained from the FDC to T cells, providing the help needed for germinal center development, production of memory cells, and the production of high affinity antibody. We will review the involvement of FDC in handling the antigen and describe these steps in some detail. In addition, we are aware of a number of these FDC dependent steps that are abnormal in aged and retrovirus infected mice and men. These abnormalities in FDC function will be discussed in the context of what is happening in young adult animals where germinal center reactions and recall antibody responses are normal. |
