Summary Shoot bud induction and their subsequent differentiation into young shoots has been observed from the cultured hypocotyl segments and embryos of Pinus wallichiana . The excised segments of hypocotyl were induced to form shoots with interphase of callus. Embryos were cultured on a modified nutrient medium along with 0.1 ppm NAA and a continuous subculturable callus was obtained. Subsequent subculturing of the callus on the same basal medium supplemented with 1 ppm BAP resulted in shoot bud formation. Embryos were also induced to form numerous shoot buds along the surfaces of their cotyledons and hypocotyls. The shoot buds when excised and transferred to a different nutrient medium, formed shoots. |
