This chapter discusses the development of an on-line glucose analyzer for bioprocess monitoring. The device consists of a pump, two 3/2-way valves, sampling probe with a sterilizable membrane and a flow cell with a biosensor for glucose determination. A continuous carrier stream is pumped through the sampling probe, located between the two valves or bypassed to the detector. Sampling is based on the principle of “stopped-flow diffusion.” By switching the valves, the flow inside the probe stops and the liquid element next to the membrane is accumulated with glucose. The amount of glucose penetrating the membrane is dependent on the stop time. With the next switching glucose is carried to the flow cell, being amperometrically detected by a glucose oxidase covered thick film electrode. First results demonstrate the dependence of glucose diffusion on stop time. Besides, the influence of temperature on diffusion and detection was investigated. Subsequently the analyzer was applied for bioprocess monitoring of E. coli fermentation. |
