Effect of Phenolic Antioxidants, BHA and BHT, on the Kinetics of Metabolic Processes of Benzo[a]pyrene and its Two Metabolites 3-Hydroxy- and 9-Hydroxy-Benzo[a]pyrene in Single Living RTG2 Cells

Popis: | The phenolic antioxidants BHA and BHT modulate the initiation and promotion of carcinogenesis by polycyclic aromatic compounds, by decreasing the binding to DNA of active metabolites. Microfluorometric studies of B(a)P metabolism in single living 3T3 have shown that in intact cellular systems BHA and BHT there is no direct MFO activity inhibition and that the study of the effect of BHA and BHT on activation and detoxication process was necessary. RTG2 cells which accumulate 3-OH and 9-OH B(a)P when they are treated with B(a)P offer this opportunity. BHA and BHT acute treatment induces an increase of the intracellular pool of B(a)P phenols due to their competition for detoxication system. Competition of the phenols and B(a)P for the MFO results in a decrease of B(a)P metabolism. 18h pretreatment by BHA or BHT does not show a significant modification of B(a)P metabolism, but it indicates a changes in the iso-enzyme pool producing 9–10 epoxides under the influence of BHA and BHA and BHT compete less... |
