Trichoderma harzianum was transformed with a plasmid p3SR2, carrying the dominant selectable marker amdS. The amdS transformants can be identified, from non-sterile soil. on Trichoderma-selective medium (TSM). containing acrylamide as the sole nitrogen source. The transformants were stable after at least 6 weeks in non-sterile soil in the greenhouse and after 14 months in sterile soil at 4°C. Soil inoculation resulted in effective colonization of the roots by the transformants. The advantages of these transformant fungi in the study of fungal ecology and population dynamics is discussed. |
