A Method for Fast Determination of Biocatalyst Process Stability

Popis: A major bottleneck in the development of industrial biocatalysts is the lack of fast and efficient methods for determining the catalyst process stability. In order to solve this problem, a new method has been developed for fast in-situ characterisation of activity and long-term stability. The theory necessary for modelling enzyme activation and deactivation is presented for the example of a first-order deactivation mechanism. As model reaction we have chosen the carboxyesterhydrolase-catalyzed hydrolysis of triglycerides, which was performed in a continuous flow reactor. To accelerate the ageing process, the temperature was continuously increased during the experiments. The procedure of parameter determination on the basis of experimental results is described. The influence of immobilisation and of the reaction media on activity and stability of the immobilised enzymes was investigated as an application of the new method. The temperature dependence of characteristics such as initial activity, half-life, and turnover number were determined.
