Use of Percolation for the Extraction of Vasopressin1

Popis: Beef posterior pituitary powder was percolated with ethanol containing increasing concentrations of water and of acetic acid. The percolate with 70% ethanol containing 0.05 M acetic acid was rich in vasopressin. Further purification of vasopressin was effected by electrophoresis in 0.5 M acetic acid at 1000 volts. The best preparation of vasopressin, produced by a combination of percolation and electrophoresis, had a potency of 450 units per mg. Acid hydrolysis of this preparation yielded only the amino acids of arginine vasopressin.
