Phytochelatin (PC) synthase of tobacco cells (Nicotiana tabacum cv. BY-2) was characterized. The enzyme was most stable in a narrow pH range around 7.5. Cd2+ and glycerol stablized this enzyme in vitro. The Km for glutathione (GSH) was 8.6 mM, which was higher than the GSH concentration in the plant cell. Both of the level of induction of cellular PC in response to Cd2+ exposure and the level of Cd-tolerance were elevated by application of exogenous GSH. This suggests that cellular GSH concentration is one of the limiting factors of PC biosynthesis and Cd-tolerance. |
