The consumption of osmium tetroxide by components of the cytoplasmic membrane of Acholeplasma laidlawii and its morphological implications

Popis: | The amounts of osmium tetroxide consumed by the double bonds of the unsaturated fatty acyl chains, by the lipid polar head groups and by proteins in the cytoplasmic membrane of Acholeplasma laidlawii were determined. Our results indicate that of the total amount of osmium tetroxide consumed, approximately 0.08 mg per mg dry membranes was consumed by the membrane proteins and the rest by the unsaturated acyl chains of the membrane lipids. The lipid polar head groups did not consume any osmium tetroxide. These results agree with the assumption that about 1 mole of osmium tetroxide is consumed per fatty acid double bond. The cytoplasmic membrane appeared trilaminar in electron micrographs of stained sections of osmium-fixed cells irrespective of the amounts of unsaturated acyl chains present in the membrane. In the absence of osmium the membrane also appeared trilaminar but slightly thicker and more diffuse. Our results, together with earlier findings, indicate that the trilaminar appearance of cytoplasmic membranes is mainly caused by membrane proteins in combination with a staining agent. |
