Autophagy dysfunction has been linked to various neurodegenerative disorders, which are characterized by the accumulation of different protein aggregates. Autophagy is known to control the expression of wild-type as well as aggregated forms of TDP-43, in vitro and in vivo. Similarly, TDP-43 is also implicated in the regulation of autophagy. It is evident that autophagy dysregulation is predominant in the development of TDP-43-associated neurodegenerative diseases, such as amyotrophic lateral sclerosis. This chapter discusses various aspects of the association between TDP-43 and autophagy in the context of neurodegenerative disorders. Overall, the regulation of autophagy has been suggested as a promising therapeutic strategy of neurodegenerative diseases. |
