Observations on the Comparative Infectivity of Cellular Fractions Derived from Homogenates of Mouse-scrapie Brain

Popis: Brain homogenates from mice clinically affected with mouse-scrapie were submitted to a simple centrifugal fractionation procedure. The infectivity of the various fractious was compared by titration in mice, and it appeared that the scrapie agent was concentrated in the particulate fractions of the cell. The highest levels of infectivity were found in the fraction rich in mitochondria and in the more readily sedimentable portion of the microsomal fraction. A slightly more refined fractionation procedure was used to show that the infectivity was unlikely to reside in myelin and that nuclei probably contained relatively small amounts of the agent. It is suggested that an assay of the mouse-scrapie agent can be made by a combination of partial titration and a measurement of incubation periods.
