Investigation of the Levels of Heavy Metal in the Blood and Venous Blood and MicroRNA Levels in Women With Migraine

Popis: In this study, it is aimed to investigation of microRNA (let-7b, miR-21, miR-22, miR-27-b, miR-34a-5p, miR-126-5p, miR-155-5p and miR-382-5p) and heavy metal levels (Hg, Mn, Co, Ni, As, Cr, Pb, Ag, Se) at the same time in the cupping blood and venous blood in female migraine patients. RNAs isolated from plasma samples by using High Pure microRNA Isolation Kit. RNA samples converted to cDNA by using miScript II RT Kit. cDNA samples are PreAmplified by using miScript Microfluidics PreAMP Kit. qRT-PCR analysis performed by using miScript microRNA Assays with Dynamic Array 96.96 on BioMark System. An ELAN DRC-e inductively coupled plasma mass spectrometry (ICP-MS) system was used for the measurements of the levels of trace elements in serum and using an multielement standard solution for IV and ICP. The MARS microwave digestion system was employed for mineralisation of serum samples. Serum total cholesterol, triglycerides, high density lipoprotein cholesterol, LDL- cholesterol and blood glucose was measured by commertially available kits based on routine methods by the Abbott Architect C16000 auto-analyzer.
