Transgenic pigs which produced from 0.1 to 1.0 mg m1−1 of recombinant human Protein C (rHPC) in milk were generated. A process for the purification of rHPC from milk has been developed which used selective precipitation by polyethylene glycol (PEG), enrichment of rHPC by barium/citrate precipitation, viral inactivation by solvent/detergent treatment and fractionation of rHPC on an ion exchange column. The overall recovery of rHPC by this process was 24–35%. rHPC was 95% pure and had enzymatic and anticoagulant activities similar to those of plasma derived HPC. These results demonstrate that transgenic pigs can be efficient “bioreactors” for the production of rHPC, and that the purification of the protein from milk by conventional methods can be developed for economical manufacture. |
