Popis: Natural product pathways are remarkable for their ability to produce medicines and other compounds with useful biological activity. Although methods such as native host cultivation and in vivo heterologous expression are powerful techniques in understanding biosynthesis, in vitro reconstitution of the pathway offers distinct advantages in elucidating precise details such as enzyme substrate specificity and intermediate steps of product formation. Here, we provide an overview of how in vitro reconstitution has been utilized to elucidate mechanisms of natural product biosynthesis and discuss its future outlook in the context of biosynthetic pathway characterization.
