The peptidoglycan of N. gonorrhoeae grown in continuous culture was labelled with [14C]- glucosamine, digested with Chalaropsis B muramidase and the products separated on TLC. The degree of O-acetylation of peptidoglycan estimated from the TLC was 25% for strain P9-2 and 34% for strain KW-2, appreciably lower than the O-acetylation of peptidoglycan isolated from batch grown cultures (47% and 57% respectively). Monomeric units (O-acetylated and non-O-acetyl-ated) were isolated by gel filtration and further analysed by reverse phase HPLC. The amount of O-acetylmonomer tetrapeptide and 1,6-anhydromuramyl residues present in the peptidoglycan was estimated. |
